That's really strange. What version of Studio One do you have, please? I tested it on Studio One 2.5 and 2.6. If you open the Editor, select One MIDI Note in the editor, and Apply the Macro, it should works for this One Note only, even if there are other MIDI Notes in the MIDI region.

I tested it again, and it wors, as expected, even if you use overlap of notes. Even if you have more notes in the track, etc.

Sounds strange... Did you try to move the event to different track, and listen to it? Just to be sure, there is (or is not) the MIDI note? Are you sure, you are using one layer only?
Could you send me (just for testing, and my own insterest) any of the project, where is this problem, please? Can you mark me, where is this problematic MIDI note (like write me down, on which track the MIDI note sounding)?
What version of Studio One do you have, now? I'm afraid, upgrade to the Studio One 2 Pro will not fix this (if you have Free or Artist or Producer version of Studio One 2). It looks like incompatibility with this sound card because of any reason. :-/
Can you try to use Agregate device of Mac OS X, and use this sound card in the aggregate device? You will bypass an internal driver of the sound card. This could works.
Can you double check your Audio Setup from Options? Check your Device Block Size (Buffer Size) is large enough. Try to increase it. You can check the Performance in the Transport bar (on the left side, there is Performance bar).
I suppose, you are using PreSonus USB ASIO driver, am I right?
kirnu arpeggiator should work proper! BUT not only kirnu, often some midi notes arent replayed as expected.
Could you be more specific, please? I don't have this problem. I have had problem with EastWest Play engine. Some notes didn't played, but this was problem of the EastWest Play engine (I tested it on different DAWs, it was exactly the same).
