Loaded up a song today and found nearly all of my audio units plugins are missing. Just to check, I looked in Garage Band and they were missing there too. I deleted the com.apple... cache files. Restarted, all au's now working fine in GB. Went in to S1, checked all folder assignments. It seemed to scan the folders and find everything when I clicked "apply". But on closing and reopening S1, no au's show up still. Any clues?

The reference Manual says something in Chapter 4.5.5. (hint to the staff: mine - german version - has 4.5.5 two times )
If S1 did not find plugins (or finds some problems with'em), it puts them on a blacklist - if you reset the blacklist, it will scan them again.

Maybe this helps.

Thanks. I had tried that a few times, but I figured I'd try again anyways... No luck. I actually watched as the scanning mentioned plugins that are missing, but when S1 opens, they are nowhere to be found.

Amazing. Seems like a system problem, if they had missed in Garageband too .

Now maybe someone else has an idea, if you post some system details.

Is S1 being run in 64-bit mode (assuming you have OSX 10.6 running this way as well, not an easy thing to do surprisingly)? If so, any AU that is not 64bit itself (most of them) will not show up, as we have no bit bridge built in.

Thanks for that, but still no go. It seems like any of these issues might have been the initial cause of the problem, but S1 still won't find them...

Have you looked them up in their folder HD/library/audio/plugins/component ? Are they there?

Other than that I really fear, without any further system details nobody will have any idea.

Although I don't really believe that this is the cause of your problems you could repair disc permissions as described in the PreSonus Knowledge Base:
